چکیده

Tyrosinase, the enzyme responsible for melanin biosynthesis in normal tissues and malignant melanomas, has been reported to be a vital respiratory enzyme of melanizing tissue. In the present study, S-91 mouse melanoma expiant growth inhibition in vitro was evaluated quantitatively for 7 substrate analogs of tyrosine, alone and in the presence of L-cysteine. i>L-j3-Phenyllactate (PLA) was the most effective inhibitor, alone and in the presence of L-cysteine. PLA alone and in the presence of Lcysteine does not inhibit the in vitro growth of hearts of new born albino mice expiants. L-Cysteine facilitates specific inhibi tion by PLA in the same manner that transient tissue chilling or transient anoxia plus PLA does, suggesting a related mecha nism. The facilitated inhibition of S-91 mouse melanoma growth by PLA plus L-cysteine in vitro suggests the possibility of such l>owerful specific inhibition in vivo.

The inhibition effects of melon on mushroom tyrosinase activity

The  inhibition effects seed and peel of melon on diphenolase activity of mashroom tyrosinase were investigated. The IC50 values and Ki values of seed and peel of melon were evaluated. increased the Km value and decreased the Vm value so it show mixed type inhibition on mushroom tyrosinase when L-DOPA was used as a substrate.

Differential Metabolic Effects of Novel Cilostamide Analogs, Methyl Carbostiryl Derivatives, on Mouse and Hyperglycemic Rat

Objective(s) PDE3 has a functional role in insulin secretion and action. We investigated the metabolic effects of new synthetic PDE3 inhibitors (mc1, mc2, mc5 and mc6), on mice and hyperglycemic rat. Materials and Methods The test compound or solvent was injected subcutaneously to mice, for 7 days. On day 8, blood and liver samples were obtained. In hyperglycemic rat, 0.5 g/kg glucose with o...

Inhibition of Helicobacter pylori Growth in vitro by Saffron (Crocus sativus L.)

Objective(s) Anti-Helicobacter pylori effects of saffron (Crocus sativus L., Iridacea) and its major constituents, crocin and safranal, were evaluated. Materials and Methods Macerated aqueous and methanol extracts tested against 45 clinical isolates of Helicobacter pylori, using paper disc diffusion method (DDM) on modified egg yolk emulsion agar (EYE agar). Four antibiotics also tested agai...

Inhibition of the growth of mouse melanoma by chlorpromazine.

The effects of three phenothiazines, chlorpromazine (CPZ), chlorpromazine sulfoxide (CPZ-O), and 74iydroxychlor promazine (7-OH-CPZ), on the growth of B-16 and HardingPassey mouse melanomas were investigated. CPZ and 7-OH-CPZ both inhibited tumor growth, but CPZ-O did not. The melanoma tyrosinase was activated by CPZ both in vivo and in vitro. There was no significant effect of CPZ on the succi...

In vitro and in silico studies of the inhibitory effects of some novel kojic acid derivatives on tyrosinase enzyme

Objective(s): Tyrosinase is a key enzyme in pigment synthesis. Overproduction of melanin in parts of the skin results in hyperpigmentation diseases. This enzyme is also responsible for the enzymatic browning in fruits and vegetables. Thus, its inhibitors are of great importance in the medical, cosmetic and agricultural fields. Materials and Methods: A series of twelve kojic acid derivatives wer...
